The AIM-listed oncology group said it had signed up three patients each to a twice-daily 120mg cohort and a once-daily 160mg cohort of AO-252, its lead treatment. All six are moving through the trial's assessment period as planned. AO-252 is designed to cross into the brain and block TACC3, a protein that helps drive genetically defined cancers. The latest step tests a new lipid-based softgel version of the drug, which the company hopes will be absorbed more predictably than the original tablet. That matters because absorption has shaped the results so far. Earlier tablet dosing produced a clinical benefit in 80% of patients taking the drug twice a day, against 40% for those taking it once daily, albeit across small groups of five. The reformulation is meant to iron out that variability and deliver steadier drug levels in the body. Coiled said it remained confident of dosing up to 30 patients on the new formulation by the end of 2026. It is also sticking to plans for initial safety and pharmacokinetic data, a measure of how the drug moves through the body, by the year end. Beyond that lies a larger phase I/II study focused on ovarian and prostate cancer.
